Lewis County is a county located in Northeast Missouri in the United States. Its county seat is Monticello. The county was organized January 2, 1833 and named for Meriwether Lewis, the explorer and Governor of the Louisiana Territory.

According to the 2000 U.S. Census, Lewis County has a total area of 510.85 square miles (1,323.1 km2), of which 505.01 square miles (1,308.0 km2) (or 98.86%) is land and 5.85 square miles (15.2 km2) (or 1.15%) is water.
